Endothelial Cell Selectivity to Nanoparticles Depends on Mechanical Phenotype

Endothelial cells (ECs) elongate in the direction of blood flow, are stiffer, and considered atheroprotective areas vasculature where flow-induced shear stress is high unidirectional softer, atherogenic, polygonal experiencing oscillatory multidirectional flow. To understand precise roles EC mechanics morphology uptake therapeutic nanoparticles (NPs) by atherogenic endothelium, human aortic ECs induced to adopt prescribed shapes imposed microcontact patterned adhesive islands. NP per cell increases with increasing spreading area decreases aspect ratio at constant area. Biomechanical analysis shows that elongated exhibit higher cellular stiffer membranes than low ratios, indicating a strong correlation between morphology, mechanical phenotype, uptake. Further, laminar endocytosed NPs far lesser extent those nonelongated chaotic, lower when cocultured same chamber. Results indicate conditions leading atherogenesis, such as low, chaotic shear-induction may be used increase preventative measure against atherosclerosis.
